Isabela and Dolores's relationship got soured when Isabela began courting Mariano.

Mirabel helped Isabela come out, everyone was supportive.
Post-Movie Isabela is very protective over Mirabel, similar to Luisa.
Isabela was only perfect so Mirabel didn't have to carry that burden. Mirabel would have to work twice as hard as anyone else in the family to impress the village or Abuela, but Isabela didn't want that for Mirabel, so she filled the spots that Mirabel couldn't.
After a while of being perfect so Mirabel didn't have to, she forgot the goal in mind, and started acting out towards Mirabel.
Post-Movie Isabela's room is much bigger, so she has room for sleepovers with the other grandkids, which they have every Friday night.
Isabela's room also didn't have the same pink and purple flowers it did before;
Now it has flowers of all different shapes and colors and several plants.
Isabela isn't close with either of her parents because she's afraid that she'd end up somehow telling them how she acts towards Mirabel, or saying rude things about her behind her back and end up in trouble with her parents.
